چکیده

امروزه استفاده از گوشت و محصولات گوشتی در اکثر کشورها در حال افزایش است.  با توجه به ارزش اقتصادی گوشت، یکی از مهم‌ترین تقلب ها در تولید فرآورده‌های گوشتی که سبب ایجاد مخاطرات بهداشتی می‌شود، استفاده از بافت‌های غیر مجاز خوراکی به جای گوشت در فرمولاسیون فرآورده است. این مطالعه با هدف بررسی و تشخیص بافت‌های غیر مجاز در فرآورده های گوشتی تجاری تولید شده توسط برخی کارخانه­های مواد غذایی اجرا شده است. برای این منظور نمونه‌ها (۳۰ نمونه ) از محصولات تجاری به صورت تصادفی جمع‌آوری شدند. هر نمونه به سه قسمت و از هر قسمت یک قطعه اخذ و برای ثبوت در فرمالین ده درصد بافر خنثی قرار داده شد. نمونه‌ها برای مطالعۀ میکروسکوپی نوری با روش‌های معمولی بافتی آماده‌سازی و در پارافین قالب‌گیری شد، و مقاطع ۶ میکرومتری از آن تهیه گردید، سپس با استفاده از هماتوکسیلین-ائوزین رنگ آمیزی شد. بررسی بافت­شناسی نشان داد که ۱۰۰ درصد نمونه‌ها دارای بافت مخطط بودند .بافت و اجزای گیاهی در ۹۳ درصد (۲۸ نمونه) مشاهده شد. بافت همبند و فاشیا در ۸۳ درصد (۲۵ نمونه) گزارش گردید. عضله صاف تنها در ۳ درصد نمونه‌ها (۱ نمونه) یافت شد. در هیچ یک از نمونه‌ها، پوست و بافت‌های ناحیه شکمی دیده نشد. بافت ریه در ۱۰ درصد نمونه‌ها (۳ نمونه)، بافت غضروف در ۲۰ درصد (۶ نمونه) و چربی در ۷۳ درصد (۲۲ نمونه) حضور داشتند.
از آنجا که روش‌های معمول کنترل کیفیت این محصولات نمی‌تواند این نوع تقلب در تولید را آشکار سازد، توصیه می‌شود آزمون‌های بافت شناسی که حساسیت و دقت بالاتری دارند، جایگزین آزمون‌های معمول شیمیایی گردد. از آنجا که حضور بافت‌های غیر مجاز در انواع فرآورده‌های گوشتی سلامت مصرف کنندگان و جامعه را تهدید می‌کند، نیاز به کنترل‌های دقیق‌تر در این زمینه توسط نهادهای نظارتی ضروری خواهد بود.

Investigation of Tissue Adulterations by Histological Method in Various Commercial Ready-to-Eat Meat Products Using Hematoxylin-Eosin (H&E) Staining

The consumption of meat and meat products has been increasing globally. Given the economic value of meat, one of the most significant forms of adulteration in processed meat products is the substitution of authorized meat with unauthorized edible tissues, which may pose serious health risks. This study aimed to investigate and identify unauthorized tissues in commercially produced meat products from selected food manufacturing companies. For this purpose, 30 samples were randomly collected from commercial products. Each sample was divided into three parts, and one piece from each part was fixed in 10% neutral buffered formalin. The samples were prepared for light microscopic examination using routine histological techniques, embedded in paraffin, sectioned at 6 micrometers, and stained with hematoxylin and eosin.
Histological analysis revealed that 100% of the samples contained striated muscle tissue. Plant tissues and components were observed in 93% (28 samples), while connective tissue and fascia were detected in 83% (25 samples). Smooth muscle was found in only 3% (1 sample). No skin or abdominal tissues were identified in any of the samples. Additionally, lung tissue was present in 10% (3 samples), cartilage in 20% (6 samples), and adipose tissue in 73% (22 samples). Since conventional quality control methods are unable to detect such adulteration, it is recommended that histological techniques, which offer higher sensitivity and accuracy, replace it with routine chemical tests. Furthermore, due to the potential health risks associated with the presence of unauthorized tissues in meat products, stricter regulatory oversight and control measures are essential.
